Abstract is a web-based network biology resource (Alexeyenko and Sonnhammer 2009; Persson et al. 2021) that is intended to help researchers in identifying and examining functionally coupled genes/proteins. The platform is a useful tool for biologists doing systems biology since it offers a user-friendly interface for interpreting and displaying the results. With the new release of FunCoup 6, we want to update the training framework and evidences, such as protein domain interactions (DOM). This project aims at integration of structural modeling of protein interactions. The remarkable abilities of AlphaFold2 to predict PPIs has already been demonstrated. Because the combination of proteins is too large to be computed with AlphaFold2, we focus on domain interactions. Once domain interactions are scored, domains will be mapped to the proteomes for 22 species. The final DOM score for proteins A and B will be a summary of the interacting domain scores. The project will integrate the new scoring system into FunCoup 6 to improve protein physical interactions. The contribution of the updated DOM to the FunCoup 6 predictions will be measured and compared to prior versions relying on UniDomInt.
