Abstract
We have developed a highly multiplexed metod for in situ mRNA detection based on single-molecule RNA in situ hybridization. The method is based on (1) optical multiplexing of five color channels (2) multiplexing by sequential hybridization and stripping. The main novelty is the high level of multiplexing, and the fact that the method works on tissue, where previous methods have only been demonstrated on cells in culture. We have demonstrated simultaneous detection of 39 genes with single-molecue resolution in mouse brain. Using this method, we now study the development of the mouse forebrain and midbrain. However the method is widely applicable, and has already been used in top publications, including two Science papers in the last year.
